In parenteral applications phospholipids especially enable the intravenous administration of poorly water-soluble drugs and can prevent precipitation of these drugs at the injection site or in the blood circulation. They serve as a viable non-toxic intravenous alternative solubilizer to synthetic detergents and organic solvents. They can encapsulate hydrophilic, lipophilic, and amphiphilic drugs, and change the body distribution pattern, which results in an improved therapeutic index of the encapsulated drug. After subcutaneous and intramuscular administration, phospholipids can ensure slow and extended release by means of encapsulation or dispersion of the drug.

Phospholipids are functional ingredients in many different types of formulations for parenteral administration.

Emulsions
The most prominent application of phospholipids in injectable formulations is as emulsifiers in O/W emulsions for parenteral nutrition. The abundant use of these systems at high dose levels, commonly based on egg phospholipids as emulsifier, demonstrates the excellent suitability of phospholipids as excipients for parenteral formulations and their unsurpassed safety profile. Phospholipid-based emulsions are essential for the delivery of oil-soluble drugs such as propofol.

Liposomes
Liposomes serve as drug carriers for improving the therapeutic index of active pharmaceutical ingredients (APIs). They are able to enrich the encapsulated drug after intravenous administration in, e.g., tumor tissue and/or to bypass organs which are sensitive to the toxicity of the drug. The residence time in blood circulation can be tuned by the composition of the liposome. Different types of depot systems for local, intramuscular and subcutaneous administration can be formed with phospholipids.

(Mixed) Micelles
Mixed Micelles – comprising phosphatidylcholine and bile salts — are suitable for solubilizing poorly water-soluble substances such as fat-soluble vitamins. These systems imitate the natural uptake mechanism of the body to ensure safe bioavailability of lipophilic actives.

Suspensions
Phospholipids added to drug suspensions in aqueous media, for subcutaneous or intramuscular administration, serve as wetting agents to increase the dispersibility of the suspended drug.

Lipid Nanoparticles
Lipid nanoparticles comprise a new platform for the delivery of RNA and other drug substances. Phospholipids act as essential building blocks and surfactants, stabilizing solid lipid particles in aqueous media, and play an important role in cellular uptake, endosomal escape, and intracellular trafficking processes.
