F-CAD Cloud brings formulation design to your desktop

F-CAD Cloud is a web software platform for in silico pharmaceutical formulation design based on the latest GPU-powered massively parallel computing paradigm. F-CAD Cloud reduces the laboratory work needed to design a new solid dosage medicinal product, such as a tablet or a capsule, replacing burdensome screening experiments and making only confirmative laboratory tests necessary. As a result, using F-CAD Cloud can accelerate time to market and reduce the risk of failure compared to traditional formulation development.

Based on the latest GPUs available in S/M/L/XL performance categories (currently NVIDIA Tesla M60, Tesla V100, Tesla T4, and A100), F-CAD Cloud allows for a high spatial resolution and calculation of the dissolution profiles and the design matrices at speeds never before possible without access to supercomputing infrastructure.

Individual formulation components are arranged within a three-dimensional matrix, which has the shape of a real tablet. All designed tablet or capsule shapes created with the F-CAD Tablet Designer module can be directly 3D-printed.

The computational matrix is designed with the F-CAD Cloud Particle Arrangement and Compression module. The PAC module’s functionality allows the creation of heterogeneous structures, i.e., granules with given granulometric composition, external and internal phases, granules consisting of different components, multilayer granules, and tablets, as well as an introduction of a coating layer. The F-CAD Cloud toolbox of the PAC module allows testing formulations with fast, unmodified release and sustained or delayed-release.

The simulation of the dissolution profile maps the physical and chemical properties of the used materials, such as solubility, particle sizes, pore sizes, etc., on the individual voxels. These voxels represent a grid on which the differential forms of advection-diffusion equations are solved. The F-CAD Cloud Dissolution Simulation module can take porous media into account by calculating the liquid transfer through the porous domain, simulating swelling and gelling of the component known to have some water-binding capacity, for example, HPMC. Tablet disintegration and the action of the disintegrant components is also considered during the simulation process. The dissolution simulation conditions can be set according to the major pharmacopeias’ requirements or customized for a research project.

The input of substance parameters is flexible; F-CAD Cloud allows some parameters to be left blank, meaning the corresponding physical effect will not be used in the final calculation of the release profile. For some of the substance properties, either an automatic assessment is carried out without the user’s input, e.g., the diffusivity, or a cubic-spline interpolation, for example, for the solubilities at different pH values.

F-CAD Cloud is in a beta test phase, so we’ll be using your feedback to improve the product and iron out any bugs which may appear. You can use F-CAD Cloud for commercial projects, but we may ask you for access to your data in order to understand and implement changes. We may also ask you to provide us with references for future customers in your role as an expert/opinion leader within the industry.
