Abstract:
Twin screw hot melt extrusion is increasingly deployed in pharmaceutical manufacturing for continuous wet and melt granulation, amorphous dispersions, controlled release and taste masking. With increasing usuage a better understanding of critical material attributes of excipients and interrelated process variables is required. This presentation will discuss in depth the desirable finctional and material attributes such as feedability/ flow, extrudability/ thermo plasticity and stability to shear and heat stress and impurity formation. Case studies will be presented highlighting excipient and process considerations a novel continuous granulation and drying system (Vector Granuformer), optimizing HME parameters to minimize impurities in an anti- retro viral formulation and designing an optimized extrusion excipient for amorphous dispersion and melt granulation.
