Disolcel® – Croscarmellose Sodium
Product Introduction
Disolcel® Croscarmellose Sodium is an internally cross-linked sodium carboxymethyl cellulose, which is used as super disintegrant. It is essentially water-insoluble, yet rapidly swelling and absorbing many times its weight in water. As the result, it provides superior tablet disintegration and aids in dissolution of the active ingredient. Compared to other disintegrants like SSG, PVPP, CCS provides better disintegration effect, and is effective at low usage level from 0.5 to 3 %.
It is widely used in pharmaceutical and dietary supplement in the form of tablets, capsules, and granules due to the excellent water uptake and rapid swelling properties.

In our research when usage level decreases to 0.25 % and 0.5%, Disolcel® from Mingtai shows faster drug dissolution rate than other disintegrants.

Benefits of CCS in formulation Containing Low Solubility API
In order to facilitate a smooth production process, it is necessary to add some hydrophobic excipients to the formulation, such as glidant, lubricant and more. These hydrophobic excipients usually have negative effects on tablet disintegration and drug dissolution.
The situation can be even worse for those API with low solubility.
With the increase of hydrophobic property in the formulation system, it becomes more difficult for the water to penetrate the tablet which will cause disintegration time to increase dramatically.
Disolcel® CCS is less sensitive to hydrophobic property of the tablet system which makes it a better disintegrant solution.
In the experiment, the hydrophobic excipient, Talc, is mixed with other hydrophilic excipients with various ratios to create systems with different hydrophobic properties.
Disintegration time of tablets containing 1% Disolcel® CCS in different ratios of Talc is similar to one another. The situation remains the same for day 0 and day 7 (40°C/75RH%). On the contrast, disintegration time of tablets containing 1% PVPP rises with the increase of hydrophobic properties in the system.
With the superior capability, Disolcel® CCS is more tolerant of rigorous systems, which makes it a better ingredient for designing a formulation with robust tablet strength, smooth production process and suitable disintegration time.
The difference of disintegration time becomes more significant after storing the examples under 40°C/75RH% condition for 7 days.
