Less known facts about Excipients: Magnesium Stearate

A trusted lubricant in solid dosage forms

Magnesium Stearate

This article is part of the “Less Known Facts About Excipients” series by Philippe Tschopp.

Magnesium Stearate: A trusted lubricant in solid dosage forms

Magnesium stearate is probably the most widely used lubricant in tablet manufacturing, yet it is also one of the most misunderstood pharmaceutical excipients.

Despite its name, magnesium stearate is not mined as a mineral. Instead, it is manufactured by combining a mineral with fatty acids obtained from natural oils and fats. The origin of these fatty acids can significantly influence regulatory, cultural and formulation considerations.

Magnesium stearate is the magnesium salt of stearic acid and palmitic acid. While the excipient is commonly referred to as “magnesium stearate,” pharmaceutical grades typically contain a mixture of magnesium salts derived from both fatty acids.¹–³

The fatty acids themselves may originate from:

  • Palm oil
  • Rapeseed oil
  • Sunflower oil
  • Soybean oil
  • Tallow (animal origin)

Today, many pharmaceutical manufacturers offer exclusively vegetable-derived grades to meet regulatory, religious and vegan requirements.

How is pharmaceutical magnesium stearate produced?

The production process consists of four main steps:

  1. Production of fatty acids Vegetable oils or animal fats are hydrolyzed or split into glycerol and free fatty acids. Stearic and palmitic acids are subsequently purified.
  2. Neutralization Purified fatty acids react with a magnesium source, typically magnesium oxide, magnesium hydroxide or magnesium carbonate.
  3. Precipitation and Washing Magnesium stearate precipitates as a fine white powder and is washed to remove residual salts and impurities.
  4. Drying and Milling The material is dried, milled and classified to achieve the desired particle size and bulk density before complying with pharmacopeial specifications.¹–⁵

What makes magnesium stearate unique?

Magnesium stearate is a hydrophobic metal soap with an exceptionally low coefficient of friction.

Its most valuable pharmaceutical characteristics include:

  • Excellent lubrication
  • Anti-adherent properties
  • Improved powder flow
  • Hydrophobic surface properties
  • High thermal stability

Only 0.25–2.0% is typically required to significantly reduce friction between the powder blend, punches and dies during tablet compression.¹

However, because of its hydrophobic nature, excessive blending or high concentrations may reduce tablet hardness, delay disintegration and slow dissolution—making magnesium stearate one of the most carefully controlled excipients during tablet manufacturing.

Pharmaceutical applications

Pharmaceutical-grade magnesium stearate is used in:

  • Tablets
  • Capsules
  • Powder blends
  • Granules
  • Direct compression formulations
  • Dry powder inhalation products¹,²

Its primary functions include:

  • Lubricant
  • Anti-adherent
  • Glidant (secondary function)
  • Processing aid

Considerations for formulators

Magnesium stearate is often described as a “critical excipient” because its functionality depends not only on its chemical composition but also on:

  • Fatty acid composition
  • Specific surface area
  • Particle morphology
  • Mixing time
  • Shear during blending

Even small changes in blending conditions can influence tablet disintegration and drug release, which is why lubricant optimization is an essential part of formulation development.

Another important consideration is raw material origin. Vegetable-derived grades are increasingly preferred for pharmaceutical products intended for global markets requiring vegetarian, vegan, halal or kosher compliance.¹–⁴

Major pharmaceutical manufacturers

Examples of established manufacturers and suppliers include:

  • Dr. Paul Lohmann
  • Roquette
  • Peter Greven GmbH & Co. KG
  • Mallinckrodt Pharmaceuticals Specialty Chemicals
  • Baerlocher GmbH
  • FACI S.p.A.
  • Valtris Specialty Chemicals

Did you know?

Although the name suggests a single chemical compound, pharmaceutical magnesium stearate is typically a mixture of magnesium stearate and magnesium palmitate. This composition is recognized by both the European Pharmacopoeia and the United States Pharmacopeia and contributes to its lubrication performance.

It is also one of the very last ingredients added before tablet compression. A few extra minutes of blending can noticeably change the final tablet properties—making magnesium stearate one of the smallest ingredients with one of the biggest impacts on manufacturing.
